So I spent most of today doing all that I could in the lab for about 6 hours. Boiling water, Micropipetting, and centrifuging. Its pretty tedious to do this for six hours, especially to 50 samples. I remember back in genetics that this process was exciting, but I think that was because it was a new process and it was only one sample that we did at a time.
This whole thing is pretty fun, but not having anyone in the lab with me is pretty lonely.
Here's what I did today:
I denatured all of the DNA by adding Chelex to all of the swabs and vortexing them to get all the cells off the swab. Then I removed the swabs from all of the tubes and transferred 500 microliters to a new tube. This tube is to be boiled for ten minutes and then placed in an ice bath for 2 minutes. I then had to centrifuge all the samples. After this, all of the DNA is in the liquid, rather than the cells and Chelex beads at the bottom of all of the tubes.
This whole process took me 5 and a half hours to complete,. I'm glad I didn't have more samples than 50, if I had 200 like I originally planned, this thing would have taken forever!
